E. coli can be frozen for indefinite long-term storage at -80C in the presence of 15% glycerol.

  1. StreakForIsolation and grow a small overnight culture from a single colony
  2. add 300 μl of sterile 50% glycerol to 700 μl of the overnight culture
  3. mix well by pipetting up and down and store at -80C


In this awesome deigsn of things you actually get an A+ for hard work. Exactly where you misplaced everybody was first in the details. As people say, details make or break the argument.. And it couldn't be much more correct here. Having said that, let me say to you what did deliver the results. Your writing is actually highly convincing which is possibly the reason why I am taking the effort to comment. I do not make it a regular habit of doing that. 2nd, while I can see a jumps in reasoning you make, I am not really sure of exactly how you appear to unite your ideas which in turn help to make the final result. For now I will subscribe to your position but trust in the future you actually connect the dots much better.
